Overview

Chlorpromazine + temozolomide is an investigational combination therapy being studied for the treatment of glioblastoma, particularly in patients with unmethylated MGMT gene promoter. Chlorpromazine is a first-generation antipsychotic that has demonstrated antitumor properties, including inhibition of cell viability, induction of polyploidy, reduction in cloning efficiency and neurosphere formation, and downregulation of stemness genes in glioblastoma cell lines. Temozolomide is an oral alkylating agent widely used as standard chemotherapy for glioblastoma. Preclinical studies have shown that chlorpromazine synergizes with temozolomide to reduce tumor cell viability and enhance cytotoxic effects. The combination is currently being evaluated in phase II clinical trials as part of the adjuvant phase following radiochemotherapy (Stupp protocol) for newly diagnosed glioblastoma[1][2][3][4][5].
