CHM-029 is a next-generation, small-molecule pan-menin inhibitor developed by CHARM Therapeutics for the treatment of acute myeloid leukemia (AML). Discovered using the proprietary DragonFold deep learning platform, which utilizes deep learning to model protein-ligand interactions, CHM-029 is designed to disrupt the interaction between the menin protein and KMT2A (also known as MLL1). This disruption restores normal gene regulation in leukemic cells. CHM-029 is specifically engineered to retain potency against known clinical resistance mutations that limit the efficacy of first-generation menin inhibitors. Pre-clinical studies have demonstrated dose-dependent tumor regression in both wild-type and mutant menin models, alongside a favorable safety profile. The drug is expected to enter first-in-human clinical trials in the second quarter of 2026.
