Overview

CHM-1101 is a first-in-class, autologous CAR-T cell therapy under development for the treatment of recurrent or progressive glioblastoma multiforme (GBM), astrocytoma, malignant glioma, and other solid tumors such as melanoma, small cell lung cancer, prostate cancer, and colorectal cancer. The therapy consists of a patient’s own T cells genetically modified to express a chimeric antigen receptor (CAR) that incorporates chlorotoxin (CLTX)—a peptide derived from scorpion venom—as the tumor-targeting domain. This CAR is fused with co-stimulatory signaling domains including CD28 and CD3ζ. CHM-1101 targets tumor cells by binding to specific markers present on GBM and other solid tumors via CLTX. It is administered through intracranial intratumoral or intraventricular routes. Developed by Chimeric Therapeutics in collaboration with City of Hope research center, CHM-1101 has shown promising preclinical activity against GBM with minimal off-tumor toxicity and is currently in Phase 1 clinical trials for glioblastoma and related indications[1][2][3][4][5][7].
