CHM-2101 is a third-generation autologous chimeric antigen receptor (CAR) T cell therapy targeting cadherin-17 (CDH17), a protein associated with poor prognosis and metastasis in gastrointestinal cancers. Developed by Chimeric Therapeutics and the University of Pennsylvania, this therapy incorporates both CD28 and 4-1BB co-stimulatory domains to enhance efficacy. It is designed for the treatment of advanced colorectal cancer, gastric cancer, and neuroendocrine tumors. Preclinical studies demonstrated that CHM-2101 could eradicate established tumors in multiple models without toxicity to normal tissues. The drug is currently being evaluated in Phase I/II clinical trials following FDA IND clearance[1][5][6][8].
