Drug intelligence / Profile preview
chMIL-38-DOTA is an unlabelled chimeric monoclonal antibody conjugate consisting of the anti-Glypican-1 (GPC-1) chimeric antibody chMIL-38 (miltuximab) conjugated to the macrocyclic chelator DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid). Developed by Minomic International and its spinout GlyTherix, chMIL-38-DOTA targets GPC-1, a cell surface proteoglycan overexpressed in various solid tumors, including prostate, pancreatic, and bladder (urothelial) cancers, while showing minimal expression in normal tissues. In clinical trials, unlabelled chMIL-38-DOTA has been administered as a pre-dose to block non-tumor binding sites and optimize tumor targeting prior to the infusion of the radiolabeled imaging agent MILGa (67Ga-DOTA-miltuximab). The conjugate is also being investigated for therapeutic applications when radiolabeled with beta-emitting isotopes such as Lutetium-177 (177Lu-DOTA-miltuximab) or alpha-emitting isotopes like Actinium-225.
