Overview

Chol-miRCtrl is a cholesterol-conjugated negative control microRNA (miRNA) mimic used in preclinical research to evaluate the specific effects of miRNA-based therapeutics. It consists of a scrambled, non-targeting double-stranded RNA sequence that lacks known biological targets in the species being studied, conjugated to a cholesterol moiety. This cholesterol conjugation facilitates systemic delivery and cellular uptake via lipoprotein-mediated pathways, mimicking the delivery mechanism of therapeutic cholesterol-conjugated miRNAs such as Chol-let-7a. In studies of hepatocellular carcinoma and other conditions, Chol-miRCtrl serves as a critical benchmark to distinguish the therapeutic activity of functional miRNAs from the potential off-target effects, hepatic toxicity, or renal injury associated with the delivery vehicle and the administration of exogenous oligonucleotides.
