This therapeutic regimen is being investigated by Wake Forest University Health Sciences for the treatment of vitamin D-deficient patients with operable triple-negative breast cancer (TNBC). It consists of high-dose oral Vitamin D3 (cholecalciferol) administered weekly for 20 weeks in combination with a standard neoadjuvant chemotherapy (NAC) schedule. The NAC component typically includes four cycles of doxorubicin and cyclophosphamide (AC) followed by twelve weekly cycles of paclitaxel, with the optional addition of carboplatin. The study (NCT03078254) explores the hypothesis that correcting vitamin D deficiency can improve the pathologic complete response (pCR) rate by leveraging the anti-proliferative and pro-apoptotic effects of vitamin D receptor (VDR) activation in breast cancer cells.
