Overview

This combination therapy consists of cholecalciferol (Vitamin D3) and metformin, being investigated for the treatment of infertility in women with polycystic ovary syndrome (PCOS) and Vitamin D deficiency. Metformin is a biguanide that improves insulin sensitivity and reduces hyperandrogenism, which are central to PCOS pathophysiology. Cholecalciferol is a seco-steroid that, upon conversion to its active form, binds to the Vitamin D receptor (VDR) to influence various metabolic and reproductive processes. The combination is hypothesized to provide superior outcomes in follicular development and hormonal balance compared to metformin alone. The Aga Khan University sponsored a randomized open-label trial to evaluate these effects, specifically looking at follicle size, endometrial thickness, and androgen levels.
