Drug intelligence / Profile preview
Cholesterol-loaded liposomes are lipid-based nanoparticles formulated to deliver exogenous cholesterol. In preclinical models of sepsis, these liposomes are used to restore depleted cholesterol levels in cell membranes, particularly in cardiomyocytes. Sepsis-induced hypocholesterolemia leads to a reduction in cardiomyocyte membrane cholesterol, which impairs beta-adrenergic receptor signaling and blunts the heart's responsiveness to catecholamines (such as dobutamine). By replenishing membrane cholesterol, cholesterol-loaded liposomes help restore adrenergic signaling and rescue myocardial function in sepsis-induced cardiomyopathy. Additionally, in the context of pneumococcal infections, these liposomes can act as decoy targets to bind and neutralize cholesterol-dependent cytolysins like pneumolysin.
