Choline alfoscerate + donepezil is a fixed-dose combination therapy used in the management of Alzheimer's disease and cognitive impairment. Choline alfoscerate (also known as alpha-glycerylphosphorylcholine or α-GPC) is a phospholipid precursor that increases acetylcholine synthesis and release in the brain, supporting cholinergic neurotransmission. Donepezil is an acetylcholinesterase inhibitor that prevents the breakdown of acetylcholine, thereby enhancing cholinergic activity. The combination aims to provide synergistic effects by both increasing acetylcholine availability (via choline alfoscerate) and reducing its degradation (via donepezil). Clinical studies such as the ASCOMALVA trial have shown that this combination can slow cognitive decline more effectively than monotherapy with either agent alone in patients with Alzheimer's disease[1][3][4][5].
