Overview

chPD1-41BB is a research-stage chimeric receptor construct designed for T-cell-based immunotherapy. It consists of the extracellular domain of the Programmed Death-1 (PD-1) receptor fused to the intracellular costimulatory domain of 4-1BB (CD137). When expressed in T-cells, this construct acts as a "chimeric switch receptor" (CSR) that converts the normally inhibitory PD-1/PD-L1 signaling axis into an activating costimulatory signal. This redirection is intended to enhance T-cell effector function, metabolic fitness, and persistence within the immunosuppressive tumor microenvironment. Preclinical studies in murine models of lymphoma and melanoma have demonstrated that T-cells engineered with chPD1-41BB exhibit a memory precursor phenotype and superior therapeutic efficacy compared to other costimulatory variants such as chPD1-CD28.
