Overview

chPD1-41BB CAR-T cells are an experimental adoptive T-cell therapy consisting of T cells engineered to express a chimeric PD-1 (chPD1) receptor. This receptor utilizes the extracellular domain of PD-1 to recognize and bind to PD-1 ligands (PD-L1 and PD-L2) expressed on tumor cells. The intracellular portion of the chimeric receptor incorporates the 4-1BB (CD137) co-stimulatory domain and the CD3 zeta signaling domain. This design aims to convert the inhibitory signals typically transmitted through the PD-1/PD-L1 axis into stimulatory signals, enhancing T-cell activation, proliferation, and persistence within the immunosuppressive tumor microenvironment. Preclinical research, including work by Medigene Immunotherapies, has demonstrated that these cells exhibit improved cytotoxic capacity, reduced exhaustion, and superior anti-tumor efficacy in models of melanoma and T-cell lymphoma.
