chPD1-CD28 CAR T cells are a preclinical chimeric antigen receptor (CAR) T cell therapy developed at Longwood University. The therapy utilizes T cells (including alpha-beta and gamma-delta T cells) engineered to express a chimeric PD-1 (chPD1) receptor. This receptor consists of the extracellular domain of the programmed cell death protein 1 (PD-1) fused to the cytoplasmic signaling domain of CD3 zeta and the CD28 costimulatory domain. By utilizing the PD-1 extracellular domain, these CAR T cells target PD-1 ligands (PD-L1 and PD-L2) expressed on various solid tumors and hematological malignancies, converting an inhibitory signal into an activating one to enhance anti-tumor efficacy. Preclinical studies have evaluated chPD1-CD28 CAR T cells in murine models of lymphoma, breast cancer, ovarian cancer, melanoma, osteosarcoma, and rhabdomyosarcoma.
