CHS-1000 is a novel, Fc-silent humanized IgG1 monoclonal antibody developed by Coherus BioSciences to target and inhibit the immunoglobulin-like transcript 4 (ILT4), also known as leukocyte immunoglobulin-like receptor subfamily B member 2 (LILRB2). ILT4 is an immunosuppressive receptor expressed on myeloid cells and enriched in tumors, where it contributes to immune evasion and resistance to PD-1 blockade. By selectively binding with high affinity to ILT4, CHS-1000 blocks its interaction with ligands HLA-A and HLA-G, reverses ILT4-mediated immunosuppression, re-polarizes suppressive M2 macrophages toward a pro-inflammatory M1 phenotype, and enhances activation of dendritic cells and T cells. This mechanism aims to overcome myeloid cell-mediated suppression in the tumor microenvironment and enhance antitumor immune responses. The drug is being developed for use both as monotherapy and in combination with other immunotherapies such as toripalimab-tpzi for advanced or metastatic solid tumors[1][3][5][8].
