CHS-114 is a fully human, afucosylated IgG1 monoclonal antibody that selectively targets chemokine receptor 8 (CCR8), which is predominantly expressed on tumor-infiltrating regulatory T cells (Tregs). By binding to CCR8, CHS-114 preferentially depletes CCR8+ Treg cells within the tumor microenvironment through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP), while sparing effector T cells and Tregs in normal tissue. This selective depletion aims to reshape the tumor microenvironment and enhance anti-tumor immune responses. The drug is being developed by Coherus BioSciences for use in advanced solid tumors, including head and neck cancer and gastric cancer. It is currently undergoing Phase 1 clinical trials both as monotherapy and in combination with toripalimab, a PD-1 inhibitor[1][2][3][4][6][7].
