**CHS-114 + toripalimab** is an investigational combination regimen composed of CHS-114, a highly selective human, afucosylated monoclonal antibody targeting the chemokine receptor CCR8, and toripalimab, a monoclonal antibody blocking the programmed cell death protein 1 (PD-1). CHS-114 is designed to selectively deplete CCR8-positive regulatory T cells (Tregs) within the tumor microenvironment, thereby enhancing anti-tumor immunity, while sparing Tregs in normal tissue. Toripalimab is an immune checkpoint inhibitor that blocks PD-1 to promote T-cell activation and antitumor responses. The combination aims to modulate both the immune microenvironment via Treg depletion and to enhance effector T-cell activity via checkpoint inhibition. This combination is being studied primarily in advanced solid tumors, including head and neck squamous cell carcinoma and gastric cancer, and is currently in phase 1 trials with demonstrated depletion of tumoral Tregs and early evidence of immune-mediated tumor responses[1][2][3][7].
