CHST15 (Carbohydrate Sulfotransferase 15), also known as GALNAC4S-6ST, is a Golgi-localized enzyme responsible for the biosynthesis of chondroitin sulfate E (CS-E), a highly sulfated glycosaminoglycan. CS-E is known to accumulate in the extracellular matrix during chronic inflammation and tissue fibrosis, and it is frequently overexpressed in the tumor microenvironment of various malignancies, including pancreatic and colorectal cancers. CHST15 antibodies are monoclonal antibodies designed to bind to and inhibit the activity of the CHST15 enzyme or neutralize its downstream effects. By reducing the production of CS-E, these antibodies aim to suppress tissue fibrosis, modulate the immune microenvironment to favor anti-tumor responses, and inhibit tumor growth and metastasis. While siRNA-based inhibitors like STNM01 have reached clinical trials, monoclonal antibodies targeting CHST15 are primarily in the preclinical research stage, being investigated for their potential in treating refractory cancers and fibrotic diseases.
