Overview

Chst15-IN-1 is a potent, reversible covalent small molecule inhibitor of carbohydrate sulfotransferase 15 (CHST15), also known as GALNAC4S-6ST. CHST15 is the specific sulfotransferase responsible for the biosynthesis of chondroitin sulfate-E (CS-E) by transferring sulfate to the C-6 position of N-acetylgalactosamine 4-sulfate. By inhibiting CHST15, Chst15-IN-1 effectively reduces CS-E sulfation levels, which can diminish the inhibitory effects of chondroitin sulfate proteoglycans (CSPGs) on axonal growth and neuronal repair. Consequently, the compound is investigated for its potential applications in stimulating recovery following central nervous system injury. Additionally, given the role of the CS-E motif in driving hormone-independent growth in castration-resistant prostate cancer (CRPC) via IL-6/STAT3 signaling, CHST15 inhibitors like Chst15-IN-1 are of interest in oncology research. It is currently available as a research chemical reagent.
