Overview

CHST15 shRNA is an RNA-based therapeutic approach designed to silence the expression of Carbohydrate Sulfotransferase 15 (CHST15), an enzyme responsible for the biosynthesis of chondroitin sulfate E. CHST15 plays a critical role in tumor matrix remodeling and the maintenance of an immunosuppressive microenvironment, particularly in refractory cancers like pancreatic ductal adenocarcinoma. By utilizing short hairpin RNA (shRNA) to knock down CHST15, this approach aims to reduce matrix-induced physical barriers and immunosuppressive signals, thereby increasing the infiltration and activation of CD4+ and CD8+ T cells within the tumor and its draining lymph nodes. Preclinical studies involving TME Therapeutics and academic partners have demonstrated that CHST15 silencing can significantly inhibit tumor growth and enhance anti-tumor immune responses.
