Overview

CI2420B (also referred to as C1420B) is a first-in-class, highly selective small molecule inhibitor of Casein Kinase 1 (CK1) isoforms alpha (α), delta (δ), and epsilon (ε). Developed by CasInvent Pharma, a spin-off from Masaryk University, the compound exhibits sub-nanomolar potency and is designed to overcome drug resistance in various malignancies. It has demonstrated significant efficacy in models of venetoclax-resistant acute myeloid leukemia (AML) and is also being investigated for chronic lymphocytic leukemia (CLL), prostate cancer, melanoma, and pancreatic ductal adenocarcinoma (PDAC). The company plans to file an Investigational New Drug (IND) application in 2026 and initiate Phase 1 clinical trials in 2027.
