Drug intelligence / Profile preview
Ciclopirox olamine leave-on solution is a topical antifungal formulation developed by Stiefel Laboratories (a GSK company). It contains the small molecule **ciclopirox olamine**, which exerts its antifungal effect by chelating polyvalent metal cations, such as Fe3+ and Al3+, which are essential cofactors for enzymes in fungal cells. This chelation leads to the inhibition of metal-dependent enzymes like cytochromes, catalase, and peroxidase, ultimately impairing fungal nutrient uptake and cell membrane integrity. The leave-on solution is specifically designed for the treatment and prophylaxis of dandruff and seborrhoeic dermatitis, particularly those associated with the yeast *Malassezia furfur*. Unlike rinse-off shampoos, this formulation is intended to remain on the skin or scalp to provide prolonged exposure to the active ingredient. It has been evaluated in Phase 1 studies to assess skin irritation potential compared to vehicle and other marketed solutions.
