Ciclopoli is a patented topical nail lacquer formulation containing 8% of the broad‑spectrum antifungal agent ciclopirox in a hydroxypropyl chitosan (HPCH) water‑soluble polymer matrix, developed to enhance transungual delivery for the treatment of onychomycosis of fingernails and toenails.[2][4][6][8] Ciclopirox exerts fungicidal or fungistatic activity by chelating polyvalent metal cations (e.g., Fe3+, Al3+), thereby inhibiting metal‑dependent fungal enzymes involved in mitochondrial electron transport, energy production, and other vital metabolic processes, with additional antibacterial and anti‑inflammatory effects.[1][5][9] The HPCH vehicle forms a film on the nail plate, improves nail penetration, and facilitates drug retention and distribution within the nail, resulting in higher ciclopirox delivery into and across the nail compared with conventional, water‑insoluble lacquers.[2][4][6]
