Drug intelligence / Profile preview
Ciclosporin gel is a localized drug delivery system developed by PerioC Limited for the treatment of chronic periodontitis. The formulation contains the immunosuppressant ciclosporin (cyclosporine A) and is intended for administration directly into the periodontal pocket following mechanical debridement (scaling and root planing). Ciclosporin functions as a calcineurin inhibitor; by binding to intracellular cyclophilin, it forms a complex that inhibits the phosphatase activity of calcineurin, thereby preventing the dephosphorylation of the nuclear factor of activated T-cells (NFAT) and subsequent transcription of pro-inflammatory cytokines such as interleukin-2. In periodontitis, this local application aims to modulate the host immune response to subgingival plaque, reducing tissue destruction and promoting healing without the systemic side effects associated with oral ciclosporin.
