ciclosporine + prednisone + rituximab

This combination therapy regimen, consisting of ciclosporine, prednisone, and rituximab, was evaluated by Nantes University Hospital as a first-line treatment for chronic graft-versus-host disease (cGVHD) in the Phase II clinical trial NCT01135641. Ciclosporine is a calcineurin inhibitor that suppresses T-cell activation, while prednisone is a corticosteroid that provides broad anti-inflammatory and immunosuppressive effects via the glucocorticoid receptor. Rituximab is a chimeric monoclonal antibody targeting the CD20 antigen on B-cells, leading to their depletion. The study aimed to determine if adding rituximab to the standard dual therapy of ciclosporine and corticosteroids could improve response rates in patients newly diagnosed with cGVHD following allogeneic stem cell transplantation.
