Drug intelligence / Profile preview
CID-016 (also known as CIR7-7321) is an orally bioavailable macrocyclic peptide developed by Circle Pharma. It acts as a first-in-class dual inhibitor of cyclins A and B, specifically targeting the conserved RxL substrate-binding motif on these cyclins to disrupt their interaction with key substrates (such as E2F and MYT1) and prevent downstream phosphorylation by cyclin-dependent kinases (CDKs). In preclinical studies, CID-016 has demonstrated potent antiproliferative and antitumor activity, inducing tumor regression in patient-derived xenograft (PDX) models of triple-negative breast cancer (TNBC), small cell lung cancer (SCLC), and ovarian cancer, particularly in tumors with dysregulation of the RB1/E2F pathway.
