Drug intelligence / Profile preview
CID755673 is a potent, selective, and cell-active small molecule inhibitor of protein kinase D (PKD). Discovered through a high-throughput screening campaign in collaboration with the University of Pittsburgh, CID755673 inhibits all three PKD isoforms (PKD1, PKD2, and PKD3) with nanomolar potency. Unlike typical kinase inhibitors, CID755673 is non-ATP-competitive, suggesting it binds to an allosteric site on the enzyme, which contributes to its high selectivity over other kinases such as PKC, Akt, and CaMKII. In vitro, CID755673 blocks PKD-mediated cellular functions, including prostate cancer cell proliferation, migration, and invasion. It has also been investigated in preclinical models for its potential to reduce the severity of acute pancreatitis and inhibit neurogenic prostate smooth muscle contractions.
