Ciforadenant is an investigational small molecule immune checkpoint inhibitor that acts as a selective and reversible antagonist of the adenosine A2A receptor (ADORA2A). By blocking this receptor on immune cells such as T lymphocytes, natural killer cells, macrophages, and dendritic cells, ciforadenant prevents tumor-derived adenosine from suppressing anti-tumor immune responses in the tumor microenvironment. This mechanism aims to restore immune surveillance and promote tumor regression. Ciforadenant is being developed primarily for cancer indications, including renal cell carcinoma and non-small cell lung cancer. The drug was originally discovered by Vernalis and is currently being developed by Corvus Pharmaceuticals in collaboration with Genentech and M. D. Anderson Cancer Center.
