Overview

**ciforadenant + daratumumab** is an investigational combination therapy being studied for treatment of relapsed or refractory multiple myeloma. Ciforadenant is a small molecule oral inhibitor of the adenosine-2A receptor (A2AR), which modulates the tumor microenvironment and enhances anti-tumor immune responses. Daratumumab is a monoclonal antibody targeting CD38, a protein expressed on myeloma cells, inducing cell death via multiple immune-mediated mechanisms. The combination is designed to leverage immune modulation (ciforadenant) and direct antibody targeting (daratumumab), potentially providing synergistic anti-myeloma effects. This regimen is currently under investigation in a Phase 1b open-label clinical trial for patients who have relapsed or refractory multiple myeloma[1][3][7].
