Overview

CIK_D332 is an autologous cytokine-induced killer (CIK) cell therapy product developed for the treatment of refractory B-cell malignancies, such as diffuse large B-cell lymphoma (DLBCL). CIK cells are a heterogeneous population of ex vivo expanded T cells that acquire natural killer (NK) cell-like properties, characterized by the co-expression of CD3 and CD56. CIK_D332 is manufactured from a patient's peripheral blood mononuclear cells (PBMCs) using a 14-day culture protocol involving interferon-gamma (IFN-γ), interleukin-2 (IL-2), and the bispecific antibody blinatumomab, which facilitates cell expansion and ensures the removal of contaminating neoplastic cells. The cells exert their antitumor effect through MHC-unrestricted lysis, primarily driven by the NKG2D receptor. In clinical practice, CIK_D332 is administered in combination with a monoclonal antibody like obinutuzumab, which redirects the CIK cells to target CD20-positive lymphoma cells through antibody-dependent cell-mediated cytotoxicity (ADCC) mediated by the CD16 receptor.
