Overview

Cilgavimab is a recombinant human IgG1κ monoclonal antibody developed for the prevention of COVID-19. It is produced in Chinese hamster ovary (CHO) cells and was originally derived from a neutralizing antibody isolated from a patient with SARS-CoV-2 infection. Cilgavimab binds to the receptor-binding domain (RBD) of the S1 subunit of the SARS-CoV-2 spike protein, inhibiting its interaction with human angiotensin-converting enzyme 2 (ACE2), thereby blocking viral entry into host cells. The antibody has been engineered with amino acid substitutions to extend its half-life and reduce effector function, minimizing potential risks such as antibody-dependent enhancement. Cilgavimab is typically administered in combination with tixagevimab as part of Evusheld for pre-exposure prophylaxis against COVID-19 in individuals at risk or who may not mount an adequate response to vaccination[2][4][6][8].
