CIM053-SG3376 is an antibody-drug conjugate (ADC) targeting CD45, also known as leukocyte common antigen. It is designed as a CD45 inhibitor for the treatment of neoplasms, particularly hematologic malignancies. The drug consists of an anti-human CD45 antibody conjugated to a cytotoxic pyrrolobenzodiazepine (PBD) dimer payload via a non-cleavable linker (SG3376). Its mechanism involves selective binding and internalization into CD45-positive cells, leading to targeted cell death while sparing non-hematopoietic tissues. This approach aims to provide safer conditioning regimens for hematopoietic stem cell transplantation and gene therapy by ablating recipient hematopoietic stem cells with reduced off-target toxicity compared to traditional chemotherapy or radiotherapy. CIM053-SG3376 was initially developed by Cimeio Therapeutics AG and remains in preclinical development[1][2][3][4][7].
