Drug intelligence / Profile preview
CIM058-tesirine (also known as CIM058-tes) is a preclinical-stage antibody-drug conjugate (ADC) developed by Cimeio Therapeutics in collaboration with the University of Basel, Basel University Hospital, and Ridgeline Discovery. It consists of CIM058, a highly potent humanized monoclonal antibody targeting CD117 (c-KIT), conjugated to the pyrrolobenzodiazepine (PBD) dimer payload tesirine. CIM058-tesirine is designed to selectively deplete CD117-expressing cells, specifically wild-type hematopoietic stem and progenitor cells (HSPCs), as a non-genotoxic conditioning regimen prior to hematopoietic stem cell transplantation (HSCT). When paired with donor HSPCs genetically engineered to express a shielded CD117 variant (such as CD117 variant 3.2 or E73K), CIM058-tesirine can eradicate host HSPCs and leukemia cells while sparing the engineered graft, enabling targeted post-transplant immunotherapy and in vivo selection.
