CIM109 is an investigational chimeric antigen receptor (CAR) natural killer (NK) cell therapy being developed by CiMaas for the treatment of solid tumors. The therapy utilizes a dual-action approach by expressing a CAR to target tumor cells while incorporating a CRISPR-mediated knockout of the TIGIT (T cell immunoreceptor with Ig and ITIM domains) checkpoint receptor. TIGIT is an inhibitory receptor expressed on NK cells that can be exploited by tumors to evade immune surveillance. By removing TIGIT, CIM109 is designed to prevent immunosuppression within the tumor microenvironment, thereby enhancing the cytotoxic activity, persistence, and anti-tumor efficacy of the CAR-NK cells against solid malignancies. It is currently in preclinical development.
