Overview

Cimetidine + promethazine hydrochloride is a combination of two small molecule drugs, each with distinct mechanisms of action. Cimetidine is a histamine H2 receptor antagonist that reduces gastric acid secretion by competitively inhibiting the action of histamine at H2 receptors on gastric parietal cells. It is primarily used for gastrointestinal disorders such as peptic ulcer disease, gastroesophageal reflux disease (GERD), and conditions involving excessive stomach acid production[6][7][10]. Promethazine hydrochloride is a first-generation antihistamine that acts as an H1 receptor antagonist. It is used to treat allergic reactions, nausea and vomiting, motion sickness, and as a sedative[3][8]. The combination has been studied for use in dermatological conditions such as atopic dermatitis and for the prevention of anaphylactoid reactions due to their complementary blockade of both H1 and H2 histamine receptors[2][4][5]. This dual blockade can provide more comprehensive inhibition of histamine-mediated effects than either agent alone.
