Overview

CIML-NK cell therapy is an investigational adoptive cellular immunotherapy developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of relapsed or refractory acute myeloid leukemia (AML). The therapy consists of donor-derived natural killer (NK) cells that are activated ex vivo with a specific cytokine cocktail comprising interleukin-12 (IL-12), interleukin-15 (IL-15), and interleukin-18 (IL-18). This brief activation induces a "memory-like" phenotype, resulting in NK cells that exhibit enhanced proliferative capacity, increased production of interferon-gamma (IFN-γ), and more potent cytotoxicity against leukemic cells compared to conventional NK cells. In clinical protocols at MSKCC, these CIML-NK cells are administered to patients following lymphodepletion and are evaluated in combination with the PD-L1 inhibitor atezolizumab and systemic IL-2 support to enhance anti-leukemic activity and overcome immune suppression.
