Overview

CIML NK cells are a form of adoptive cell therapy using natural killer (NK) cells that have been pre-activated ex vivo with a combination of interleukin-12 (IL-12), interleukin-15 (IL-15), and interleukin-18 (IL-18). This brief cytokine stimulation induces a "memory-like" phenotype in the NK cells, resulting in enhanced effector functions such as increased interferon-gamma production and cytotoxicity upon restimulation. These CIML NK cells exhibit transcriptional, epigenetic, and metabolic reprogramming that enables them to persist longer in vivo and display improved anti-tumor activity compared to conventional NK cell therapies. They are being investigated primarily for the treatment of hematologic malignancies such as acute myeloid leukemia (AML), myelodysplastic syndrome (MDS), myeloproliferative neoplasms, as well as solid tumors including renal cell carcinoma and urothelial carcinoma. Clinical trials have demonstrated their safety profile and potential efficacy when used alone or in combination with other agents like low-dose IL-2 or radiotherapy[1][2][3][4][5][6].
