Overview

CIML-NK cells + atezolizumab is an investigational combination immunotherapy regimen being developed by Memorial Sloan Kettering Cancer Center for the treatment of relapsed or refractory acute myeloid leukemia (AML). The therapy consists of allogeneic natural killer (NK) cells that are pre-activated ex vivo with a cytokine cocktail of IL-12, IL-15, and IL-18 to induce a 'memory-like' phenotype. These cytokine-induced memory-like (CIML) NK cells exhibit enhanced effector functions, including increased interferon-gamma (IFN-γ) production and potent cytotoxicity against leukemia cells, along with improved in vivo persistence. The regimen combines these cells with atezolizumab, a monoclonal antibody targeting programmed cell death 1 ligand 1 (PD-L1), to prevent immune checkpoint-mediated suppression of the NK cells by the tumor microenvironment. Additionally, low-dose recombinant human IL-2 (aldesleukin) is administered subcutaneously to support the expansion and survival of the infused NK cells.
