Overview

CIN-109 is a novel, long-acting, first-in-class growth differentiation factor 15 (GDF-15) analog developed for the treatment of obesity. It is designed to decrease appetite, maintain energy expenditure, and drive weight loss while preserving lean body mass. As a small molecule drug targeting GDF15, it acts as a modulator of this protein involved in metabolic regulation. In Phase 1 multiple ascending dose studies in obese but otherwise healthy participants, CIN-109 demonstrated good tolerability and produced meaningful weight loss—up to 3.7% within one to two months—with most of the lost weight being fat mass and preservation or increase of lean mass. The drug is administered subcutaneously either weekly or every other week; biweekly dosing was better tolerated with no serious adverse events reported[1][3][4][5][6][8][10].
