Overview

CIN-110 is a novel, long-acting analog of peptide YY (PYY 3-36), an endogenous gut hormone that activates the neuropeptide Y2 receptor (Y2R) to reduce appetite and food intake. It is being developed as both a monotherapy and in combination for the treatment of obesity. Unlike previous PYY-based therapeutics, CIN-110 is engineered to minimize severe gastrointestinal side effects such as nausea and emesis while maintaining efficacy for long-term weight loss. The drug selectively binds and activates neuropeptide Y2 receptor (Y2R), leading to reduced caloric intake, body weight loss, improved glucose homeostasis, and enhanced insulin sensitivity in preclinical models. Clinical studies have shown that CIN-110 is well-tolerated with only mild transient gastrointestinal side effects reported; it features a gradual pharmacokinetic profile with an approximate 14-day half-life following subcutaneous administration[1][2][4][5][7].
