Cinchonidine is a natural alkaloid found in the bark of Cinchona officinalis and Gongronema latifolium. It is a stereoisomer of cinchonine and is structurally related to other Cinchona alkaloids such as quinine and quinidine. Historically, cinchonidine has been used as an antimalarial agent, particularly for malaria caused by Plasmodium falciparum, often as part of combination therapies like Quinimax. In addition to its antimalarial properties, cinchonidine serves important roles in pharmaceutical development as an intermediate in the synthesis of various drugs and as a chiral ligand in asymmetric synthesis for producing enantiomerically pure compounds. Recent research suggests that it may alleviate cisplatin-induced ototoxicity by regulating PI3K-AKT signaling pathways[8]. Its mechanism of action against malaria is presumed similar to other Cinchona alkaloids—interfering with heme detoxification within the parasite—but specific molecular targets are not fully characterized[1][3][5][8].
