Cinnabarinic acid is an endogenous metabolite in the kynurenine pathway of tryptophan metabolism. It acts as a specific orthosteric agonist of the metabotropic glutamate receptor 4 (mGlu4), with no activity at other mGlu receptors. Cinnabarinic acid also functions as an agonist for the aryl hydrocarbon receptor (AhR). It has been shown to induce apoptosis and modulate immune tolerance by influencing mitochondrial respiration and neuroinflammation. Preclinical studies indicate that cinnabarinic acid exhibits analgesic effects in models of inflammatory and neuropathic pain via mGlu4 activation, and it can protect against metabolic dysfunction-associated steatohepatitis through AhR-dependent signaling pathways that regulate hepatic lipid metabolism[1][3][4][5][7].
