Overview

CInQ-03 is a novel, small-molecule, ATP-competitive inhibitor of mitogen-activated protein kinase kinase 1 and 2 (MEK1/2). Identified through research at The Hormel Institute, it targets the RAS/RAF/MEK/ERK signaling axis, a pathway frequently dysregulated in various malignancies, including colorectal cancer. Unlike many clinically approved MEK inhibitors that function as allosteric modulators, CInQ-03 competes directly with ATP for binding to the MEK1/2 enzymes. Preclinical studies have demonstrated that CInQ-03 effectively suppresses MEK1/2 kinase activity, leading to a significant reduction in the phosphorylation of downstream targets such as ERK and ribosomal S6 kinases. In vivo experiments using colon cancer xenograft mouse models showed that CInQ-03 inhibits tumor growth without inducing the skin toxicity commonly associated with this class of drugs, suggesting a potentially improved safety profile for future therapeutic applications.
