Overview

Cintredekin besudotox is a recombinant chimeric protein developed as a specific tumor-targeting agent for the treatment of malignant gliomas, particularly glioblastoma multiforme (GBM). It consists of human interleukin-13 (IL-13), an immunoregulatory cytokine, linked to a truncated and mutated form of Pseudomonas exotoxin A (PE38QQR). The IL-13 portion binds to IL-13 receptors overexpressed on tumor cells but minimally present on normal brain tissue. Upon binding, the toxin is internalized and induces apoptosis in tumor cells. Cintredekin besudotox is administered directly into brain tissue at risk for residual infiltrating GBM after tumor resection using convection-enhanced delivery (CED), which allows high local concentrations while minimizing systemic exposure[1][2][4].
