**cipargamin + INE963** is a combination therapy under clinical investigation for the treatment of malaria. Cipargamin (KAE609, NITD609) is a spiroindolone-class antimalarial targeting the Plasmodium falciparum ATPase 4 (PfATP4), a sodium (Na+) pump, thereby disrupting ionic homeostasis within the parasite. It has shown rapid parasite clearance and is being developed for both uncomplicated and severe malaria. INE963 is a novel imidazothiadiazole (ITD) derivative with potent activity against blood-stage Plasmodium falciparum, rapid kill kinetics similar to artemisinins, high selectivity for the parasite over human kinases, a long half-life, and a high barrier to resistance. The combined use of cipargamin + INE963 is aimed at achieving curative, single-dose regimens for malaria, and may address resistance mechanisms seen with single-agent therapies[1][2][4][5].
