Ciprofloxacin mannich base (CMB) is a newly synthesized small molecule derivative of the antibiotic ciprofloxacin, primarily investigated for its anticancer properties. It demonstrates cytotoxic and pro-apoptotic effects in various cancer cell lines, including lung adenocarcinoma and high-grade serous ovarian cancer. Its mechanism of action involves attenuating the MAPK signaling pathway, inducing apoptosis, and causing S phase cell cycle arrest. CMB also modulates gene expression by up-regulating p53, p21, and Bax, while down-regulating Bcl2. Additionally, it exhibits weaker inhibition of topoisomerase II. Some ciprofloxacin Mannich bases have also shown antibacterial, antifungal, and anthelmintic activities, and can be designed to improve the physicochemical properties of the parent drug, such as solubility and dissolution rate.
