ciprofol + esketamine is a combination anesthetic regimen consisting of ciprofol (HSK3486), a novel GABA_A receptor agonist, and esketamine, the S-enantiomer of ketamine which acts as an NMDA receptor antagonist. Ciprofol is a 2,6-disubstituted phenol derivative developed by Haisco Pharmaceutical Group as a more potent alternative to propofol with a potentially improved safety profile, including reduced respiratory and cardiovascular depression. Esketamine provides analgesic and sedative effects while maintaining sympathetic tone. The combination is being evaluated in clinical trials, such as NCT05861427, for anesthesia induction and maintenance in elderly patients undergoing procedures like endoscopic retrograde cholangiopancreatography (ERCP) to minimize intraoperative hypotension and hypoxemia.
