CircFam53B-219aa DC vaccine is a personalized dendritic cell (DC) vaccine loaded with circular RNA encoding the cryptic peptide CircFAM53B-219aa. The circRNA is derived from the FAM53B gene and encodes a peptide specifically expressed in breast cancer tissues, particularly those that are HER2-negative. The vaccine is designed to activate antigen-specific T cell immune responses by presenting the tumor-associated antigen via autologous dendritic cells, thereby stimulating an anti-tumor immune response. Preclinical studies have shown that this approach can significantly inhibit tumor growth in models with high CircFAM53B expression. It is currently being evaluated as monotherapy and in combination with camrelizumab (a PD-1 inhibitor) for patients with HER2-negative advanced breast cancer who have failed first-line treatment[1][2][3][4].
