circIL-15 refers to a therapeutic approach using circular RNA (circRNA) technology to encode and express the Interleukin-15 (IL-15) cytokine. Unlike linear mRNA, circular RNA lacks 5' and 3' ends, which protects it from exonuclease-mediated degradation, potentially leading to more stable and prolonged protein expression. In the context of oncology, circIL-15 is typically delivered via lipid nanoparticles (LNPs) to the tumor microenvironment or systemic circulation, where it induces the production of IL-15. This cytokine then promotes the proliferation and activation of natural killer (NK) cells and CD8+ T cells, enhancing the anti-tumor immune response. Several biotechnology companies, including Therorna and Orna Therapeutics, are developing circular RNA platforms that include IL-15 as a primary or adjunctive payload.
