Circle Pharma's Cyclin E program is a research-stage drug discovery effort focused on developing oral macrocycles that target Cyclin E. Utilizing the company's proprietary MXMO™ platform, these macrocycles are designed to disrupt the protein-to-protein interaction (PPI) between the cyclin's hydrophobic patch and the RxL motif of its substrate proteins. Cyclin E is frequently upregulated in various malignancies, including uterine and ovarian cancers, and its overexpression is a recognized mechanism of resistance to CDK4/6 inhibitors. By inhibiting Cyclin E-mediated signaling through this non-catalytic site disruption, the program aims to address unmet needs in solid tumors where Cyclin E drives aberrant cell cycle progression and therapeutic resistance.
