CircMab is a circular RNA (circRNA) platform developed by Therorna Inc. for the in vivo production of therapeutic proteins, including antibodies and bispecific T-cell engagers (BiTEs). The platform utilizes a proprietary splint-free enzymatic circularization technology and a targeted lipid nanoparticle (LNP) delivery system. By leveraging the stability and durable protein expression characteristic of circular RNA, CircMab is designed to provide sustained therapeutic levels of proteins in the body, potentially overcoming the half-life limitations of traditional recombinant protein infusions. A primary application of the platform is a circular RNA-encoded CD19×CD3 T-cell engager intended for deep B-cell depletion in the treatment of autoimmune diseases.
